This antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 containing 5.0% w/v trehalose with no calcium, magnesium, or preservatives present.

Storage and Handling
The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 0.2-0.8 µg/ml. A suitable detection antibody is PN:S118 at a concentration of approximately 0.1-0.4 µg/ml. A suggested standard for this assay is PN:S112.
Western Blotting: To detect Mouse SCF this polyclonal antibody can be used at a concentration of 0.1 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:S112.

Neutralization: Measured by its ability to neutralize SCF/c‑kit Ligand-induced proliferation in the TF‑1 human erythroleukemic cell line [Kitamura, T. et al. (1989) J. Cell Physiol. 140:323]. The Neutralization Dose (ND50) is typically 0.3-0.6 µg/mL in the presence of 25 ng/mL Recombinant Mouse SCF/c‑kit Ligand.

Background
Stem cell factor (SCF), otherwise known as KIT ligand or Steel factor1 is a paracrine regulator of germ cell development.2 SCF has the ability to stimulate the growth of hematopoietic progenitor cells and to generate colony-forming cells (CFC) from highly enriched populations of hematopoietic cells.3 SCF is produced by the Sertoli cells and involved in the local regulation of spermatogenesis4 and liver regeneration.5 SCF in conjunction with interleukin-31 may play a significant role in airway remodeling by promoting the recruitment of bone marrow-derived fibroblast precursors into the lung with the capacity to promote lung myofibroblast differentiation.6
