DescriptionBackground Fixation Buffer is useful for intracellular staining procedures in preparation of cells for staining intracellular cytokines or other proteins. Fixation Buffer is used to fix cells prior to permeabilization using Permeabilization Wash Buffer (10X) (Leinco Product No.: K144). Leinco Technologies fixation Buffer has been formulated with prescreened paraformaldehyde with low background, thus producing the greatest signal to noise ratio. Directions for Use For cell fixation or if fixing is desired, add 0.5 ml of the working concentration 1X fixative solution per tube and leave it in the dark for 20 minutes at room temperature. Vortex the cells gently and they are now ready for flow cytometric analysis. Do not fix cells if they are to be used for tissue culture. Fixed cells should be stored at 4oC and are now stable up to 48 hours for analysis. Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change. Hazard Description Caution: This buffer contains paraformaldehyde, which is toxigenic and mutagenic. Please handle with caution and wear gloves, lab coat and necessary protection to avoid direct body contacts.
