Background The human germinal-center-associated lymphoma (HGAL) gene was originally identified from an expressed sequence tag that was associated with improved survival in patients with diffuse large B-cell lymphoma (DLBCL). Cloning and characterization of HGAL indicated that it is induced by IL-4. HGAL is a cytoplasmic protein and has been demonstrated in follicular lymphoma, Burkitt lymphoma, lymphocyte-predominant Hodgkin lymphoma, and a subset of DLBCL. HGAL expression is an important predictor of overall survival in patients with germinal center- derived tumors. HGAL protein is also found in germinal centers of normal tonsils and lymph nodes. Function Involved in the negative regulation of lymphocyte motility. It mediates the migration-inhibitory effects of IL6. Serves as a positive regulator of the RhoA signaling pathway. Enhancement of RhoA activation results in inhibition of lymphocyte and lymphoma cell motility by activation of its downstream effector ROCK.
