Anti-Human Fractalkine

Purified Recombinant Human Fractalkine (>98%)

This antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.3 containing 5.0% w/v trehalose with no calcium, magnesium, or preservatives present.

Lyophilized

The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Neutralization: This antibody is useful for neutralization of Human FKN bioactivity. The antibody dose required to neutralize 50% (ND₅₀) of the biological activity of Human FKN (at 20 ng/ml) is 0.3 - 5 µg/ml.

Fractalkine (FKN) is a transmembrane mucin-chemokine hybrid molecule expressed on activated endothelium¹ that mediates attachment and firm adhesion of T cells, monocytes and NK cells.² FKN plays an important proinflammatory role in rheumatoid arthritis (RA) pathogenesis as characterized by induction of synovial angiogenesis, chemotaxis, activation of monocytes and T cells as well as the stimulation of proliferation and synthesis of matrix degrading enzymes (matrix metalloproteinases, MMP) in synovial fibroblasts. Fractalkine thus may represent a novel target molecule for therapeutic intervention in RA.³ FKN is also an essential biomarker for predicting the prognosis of patients with colorectal cancer (CRC).⁴
