Antibody DetailsProduct DetailsReactive Species Human Host Species Goat Immunogen Purified Recombinant Human IL-3 (>98%) Endotoxin Level <0.1 EU/µg as determined by the LAL method Formulation This antigen affinity purified polyclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco’s phosphate buffered saline (1X PBS) pH 7.2 – 7.4 containing 5.0% w/v trehalose with no calcium, magnesium, or preservatives present. State of Matter Lyophilized Storage and Handling The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months. Country of Origin USA Shipping Next Day Ambient RRIDAB_2830541 Applications and Recommended Usage? Quality Tested by Leinco Western Blot: Suitable for use at concentration of 0.1 µg/mL. Additional Applications Reported In Literature ? Immunocytochemistry: Suitable for use at concentration of 5-15 µg/mL. Neutralization: Measured by its ability to neutralize IL‑3-induced proliferation in the TF‑1 human erythroleukemic cell line. Kitamura, T. et al. (1989) J. Cell Physiol. 140:323. The Neutralization Dose (ND50) is typically 0.05-0.25 µg/mL in the presence of 1.25 ng/mL Recombinant Human IL‑3. Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change. DescriptionDescriptionSpecificity Anti-Human Interleukin-3 (IL-3) recognizes Human IL-3. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research. Background IL-3 is a 17 kD pleiotropic cytokine and a member of a family of growth factors that sustains both the proliferation and the development of hematopoietic precursors. It also displays neurotrophic activity and is thought to be associated with neurologic disorders. Signal transmission is assured by high affinity binding to the IL-3 receptor (IL3RA). The receptor contains α and β subunits. IL-3 shares the β subunit with IL-5 and GM-CSF which explains their biological functional similarities. The IL-3/receptor complex induces JAK2/STAT5 cell signalization pathway and it can stimulate both the activation of gene expression and the suppression of apoptosis via transcription factor c‑myc and the Ras pathway, respectively. IL-3 fuels the differentiation of multipotent hematopoietic stem cells into myeloid progenitor cells or lymphoid progenitor cells (when in conjunction with IL-7). Together with other cytokines, including EPO, GM-CSF, and IL-6, IL-3 can stimulate proliferation of all cells in the myeloid lineage. The constitutive expression of IL-3 in some leukemia cell lines (as opposed to selective T-cell expression in response to specific impulses or antigens) is thought to fuel the development of the disease. Post-chemotherapy IL-3 application can promote regeneration of granulocytes and platelets. Furthermore, during high-dose chemotherapy, the combination of IL-3, GM-CSF and stem cell factor increases peripheral blood stem cells. IL-3 is thought to have therapeutic potential in lymphohematopoietic disorders and solid cancers. PubMed NCBI Gene Bank ID UniProt.org References & Citations1. Laston, D. et al.
