Antigen Distribution β-catenin 1 is widely expressed in many tissues, including kidney, lung,
adult and developing central nervous system, several hair follicle cell types, colon, cortical
neurons, and various cancers. Background β-catenin is an armadillo protein that binds to numerous partners, including cadherins,
adenomatosis polyposis coli, and T-cell factor 1 , and is also a principal component of the Wnt signaling pathway 2 . β-catenin is necessary for embryonic development, organ differentiation and development, bone homeostasis, mammalian hair growth, and adult tissue regeneration. Additionally, β-catenin functions in cell adhesion as part of a cadherin/catenin complex, plays an important role in the structural integrity of squamous epithelial cells 3 , and is involved in synaptic plasticity 1 . β-catenin localizes to the cell cytoplasm, and once activated, translocates to the nucleus, where it activates target genes via TCF/LEF transcription factors 12F7 was generated by immunizing Balb/c mice with a purified in-frame fusion protein of β- catenin bound to maltose binding protein 4 . The β-catenin was derived from a 2.8 kb cDNA clone isolated from a chicken embryo cDNA library, of which a 2.4 kb section encoding residue 92 through the carboxy terminus was used. Half the splenic lymphocytes were fused with the P3-X63Ag8.653 myeloma cell line and the other half with Sp2/0-Ag14. The 12F7 binding epitope was mapped to a region between residues 112 and 134 via a deletion strategy. 12F7 does not cross-react with plakoglobin.
