Storage and Handling
The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Flow Cytometry: It is recommended to use the indirect method for signal enhancement when enumerating cells expressing TRAIL R2. A suggested method would be to stain cells expressing TRAIL R2 with 0.25 µg per 1 x 106 cells in a total staining volume of 100 µl followed by PN:G641.
Western Blotting: To detect Mouse TRAIL R2 this polyclonal antibody can be used at a concentration of 0.1 - 0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T263. The detection limit for Mouse TRAIL R2 is 2 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.

Neutralization: This antibody is useful for neutralization of Mouse TRAIL R2 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Mouse TRAIL R2 (at 12 ng/ml) is 0.4 - 1.2 µg/ml.

Tumor necrosis factor-related apoptosis-inducing ligand Receptor 2 (TRAIL-R2) is a cell-surface receptor involved in tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-induced cell-death signaling.1 The death ligand TRAIL bears high potential as a new anticancer agent, as binding to the death receptors TRAIL-R1 or TRAIL-R2 triggers apoptosis in most cancer cells.2 TRAIL-R2 has been shown to be associated with a decrease in the survival rates of breast cancer patients.3
