The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.

Western Blotting: To detect Rat CCL20 this polyclonal antibody can be used at a concentration of 0.1 - 0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The detection limit for Rat CCL20 is 5 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.

Neutralization: Measured by its ability to neutralize CCL20/MIP‑3 alpha -induced chemotaxis in the BaF3 mouse pro‑B cell line transfected with human CCR6. The Neutralization Dose (ND50) is typically 30-75 µg/mL in the presence of 5 µg/mL Recombinant Rat CCL20/MIP‑3 alpha.

This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
Background
Macrophage Inflammatory Protein-3 Alpha (MIP3A) also known as CCL20 or liver activation regulated chemokine (LARC) is a small cytokine belonging to the CC chemokine family. It is strongly chemotactic for lymphocytes and weakly attracts neutrophils.1 MIP3A is specifically expressed by the follicle-associated epithelia (FAE) covering intestinal Peyer's patches (PPs) and is the only known chemokine ligand for the chemokine receptor CCR6.2 MIP3-alpha is produced by intestinal epithelial cells (including cells associated with mucosal lymphoid tissues), by keratinocytes in inflamed skin, and by cells in liver and lung.3,4 It plays an important role in the inflammatory response of blood monocytes and tissue macrophages.
