Background SMRT (Silencing Mediator for Retinoid and Thyroid receptors) is a 168 kD nuclear protein. Nuclear hormone receptors are transcription factors regulated by ligand binding. In the absence of ligand, there is a physical interaction between the nuclear receptors that do not interact with Hsp90 and a family of corepressors which includes SMRT. SMRT helps target histone deacetylases and other components of a multiprotein repressor complex to the specific target promoter to silence gene transcription. Function Transcriptional corepressor (PubMed:20812024). Mediates the transcriptional repression activity of some nuclear receptors by promoting chromatin condensation, thus preventing access of the basal transcription. Isoform 1 and isoform 4 have different affinities for different nuclear receptors. Involved in the regulation BCL6-dependent of the germinal center (GC) reactions, mainly through the control of the GC B-cells proliferation and survival.
