Western blotting (WB), also called protein immunoblotting, is a technique used to detect a specific protein in a sample based on the binding of a specific antibody. In a WB, proteins are first separated by size using gel electrophoresis. Following transfer to a membrane, a primary antibody identifies the target protein. A secondary antibody conjugated to a dye or enzyme is used for detection if the primary antibody is unlabeled.
